Creating and Pasting New Sequences

Access the Library and File Tab

In order to create a new file via copy pasting the sequence, you need to access the “Library” tab or the “File” tab in the top banner. In both menus you will see the button called “New sequence” when selected it will open the new files modal box.

Select a Destination and Paste the Sequence

Give the file a name and select the destination folder for the new file. Then paste your sequence into the “Sequence” box. Please note that all nucleotide letters according to the International Union of Pure and Applied Chemistry (IUPAC) ambiguity code are accepted.

If incorrect characters, spaces or other punctuation have been included you will be notified. You can then select the “clean sequence” button to remove incorrect characters automatically from your pasted sequence.

Select Topology Settings

Once you have pasted and cleaned your desired sequence, you can select your sequence parameters. First select if the sequence is linear or circular, then select if it is single or double stranded.

Use Detect Common Features on New Sequence

If you would like to detect common features that are part of your sequence you can do so during file creation by using the “Detect common features” toggle. When this is turned on the sequence will be scanned and labelled with existing features. You will receive a “Features detected” overview where you can select which features to include in your file using the checkboxes next to the feature names.

Transferring Primers, Features and Guides when Pasting

If you are pasting from another file, into the “Sequence” box, Photo51 will automatically transfer existing primers, features and guides in you selection. You will receive a notification indicating how many items were found. When you continue you can select which attachments to keep in the new file. Files that are pasted in will have a green clipboard indicator next to them, features that were detected will not.
