Ensure your Sequence is Linear
Open the DNA Ends Modal

Ensure your Sequence is Linear

In order to modify DNA ends, you must have a linear sequence. To do this use the “Linearize” function found under the “Edit” tab of the top banner. Here you can linearize via restriction digest or PCR. Once you have linearized your sequence you are ready to modify the DNA ends.

Open the DNA Ends Modal

Navigate to “Edit” > “DNA Ends” in the top banner, this will open the DNA end editing modal box.

In the modal you will see a split view of both ends of the sequence. For each of the ends you can add 5’ phosphorylation which will be indicated with a blue P in on the corresponding strand. For each end you also have the option to select between a 5’ and 3’ overhang and how many bases should be added.
