  1. Manage Enzymes:

You can now create custom restriction enzymes lists. Navigate to the Enzymes tab, select the drop down and choose “Manage enzymes”. Choose “+ Add group” then select “Edit”. You can give your enzyme group a name and add enzymes to this list. Once finished hit the “Save” button at the top. The new group will be added to the “Enzymes” dropdown menu and you can now display your enzymes on your plasmid.

  1. Linear Ligation:

It is now possible to ligate linear fragments using Gibson assembly or Restriction cloning. Navigate to the cloning tab and select your preferred method. In the cloning modal, select the option “Continue without vector” then you can select your inserts and assemble.

  1. Virtual Gel:

The layout and export options for virtual gel have been updated. Navigate to the “PCR” tab and select “Virtual gel”. You can add lanes select files to add to lanes and simulate the gel electrophoresis for both PCR and restriction digest products. When you have finished you can export the file (PNG, JPG, PDF) or save the gel back to your library.

  1. PCR Mutagenesis:

We have added the ability to perform PCR mutagenesis. First select the region in your sequence where you want to add a mutation. Then navigate to the “PCR” tab and select “Mutagenic PCR” this will open the PCR mutagenesis modal box. Here you can introduce the desired mutation. Once you select “Create” the product of your PCR mutagenesis will be saved to your library with the mutagenic primer and mutations marked in red.

  1. Sequence Alignment

Sequence alignment is now available in the cloud version of Photo51! We have also moved alignments to a dedicated tab in the top menu. Simply navigate to the “Alignment” tab and select between “Align to reference”, “Pairwise alignment”, “Multiple sequence alignment” or “Contig assembly”

  1. Zoom:

We have enhanced zoom in the map view to make it more effective for exploring your sequence. To check out the improvements navigate to the map view in the bottom navigation banner. Then select the magnifying glass icon on the bottom right. You can then zoom into the plasmid by dragging the slider or pressing the plus and minus icons.

  1. Sequence Search:

To search your sequence (for DNA / amino acid sequences, enzymes, primers, features or guide RNAs) navigate to the sequence view with the bottom navigation ribbon. Then use ctrl + F to open the search panel then simply type in the sequence or name that you are looking for and it will be highlighted in the sequence.

  1. Feature Display:

We have now added directionality to features in the sequence view just as they appear in the map view. This should make it clear whether the feature annotation refers to the forward or reverse strand.
